Human Dental Pulp Stem Cell Osteogenic Differentiation Seeded on Equine Bone Block with Graphene and Melatonin

چکیده
Equine bone blocks have osteogenic effects promoting regeneration with biocompatibility and osteoconductivity capacity. Human dental pulp stem cells (hDPSCs) can differentiate into osteoblasts enhancing biomineralization such scaffolds. Melatonin is able to improve health mediate formation. Collagenated equine were coated ammonia-functionalized graphene-oxide (G-N) at two different concentrations (2 μg/mL, G-N2; 10 G-N10). The homogeneity of G-N coating was checked by Raman spectroscopy, whereas thermogravimetric analysis (TGA) allowed us quantify the amount deposited on blocks. aim this study investigate in vitro effect G-N-coated collagenated proliferation differentiation hDPSCs addition a melatonin. This evaluation determined after 7, 14, 21 days culture expression specific microRNAs, RUNX2 SMAD5 gene expression, osteocalcin levels, histological analysis. results showed that G-N2 G-N10 melatonin gave an optimal cell adhesion as shown analysis, increase potential confirmed microRNA levels. suggests promote process.

Stem cells from human dental pulp have been considered as an alternative source of adult stem cells in tissue engineering because of their potential to differentiate into multiple cell lineages. Recently, polysaccharide based hydrogels have become especially attractive as matrices for the repair and regeneration of a wide variety of tissues and organs. The incorporation of inorganic minerals as...
متن کاملThe Effect of Human Platelet-Rich Plasma on Adipose-Derived Stem Cell Proliferation and Osteogenic Differentiation
Background: The cultured mesenchymal stem cells (MSC) have been used in many clinical trials however, there are still some concerns about the cultural conditions. One concern is related to the use of FBS as a widely used xenogeneic supplement in the culture system. Human platelet-rich plasma (hPRP) is a candidate replacement for FBS. In this study, the effect of hPRP on MSC proliferation and os...

Background and aim: Cell-therapy is an important science because of using to treatment of critical-sized bone defects. Recent studies in this field suggest that human endometrial derived stem cells can be a great source. On the other hand, graphene and its derivatives, mainly graphene quantum dots (GQDs) have recently attracted much attention as effective factors in differentiating stem cells t...
متن کاملHuman Serum Promotes Osteogenic Differentiation of Human Dental Pulp Stem Cells In Vitro and In Vivo
Human dental pulp is a promising alternative source of stem cells for cell-based tissue engineering in regenerative medicine, for the easily recruitment with low invasivity for the patient and for the self-renewal and differentiation potential of cells. So far, in vitro culture of mesenchymal stem cells is usually based on supplementing culture and differentiation media with foetal calf serum (...
